MPP-hPER1融合蛋白穿膜的实验研究

摘要 本课题研究了MPP-hPER1融合蛋白对多种细胞的穿膜性。在分析MPPs家族分子组成特点的基础上,合成了PLNG活性肽。并且应用重组技术构建了MPP-hPER1融合蛋白表达体系,通过细胞培养、转染、免疫荧光染色等手段,观察了不同作用时间、不同温度、不同细胞系的条件下,PLNG及MPP-hPER1融合蛋白对细胞膜的穿透能力。结果显示:PLNG及MPP-hPER1融合蛋白穿透细胞膜是非能量依赖型、非时间依赖型和非细胞种系依赖型。提示生物体中有一类特殊的生物大分子,按照非经典途径进入细胞发挥生物学活性作用。实验结果提示,PLNG可以作为一种特殊的生物分子载体,介导功能分子进入细胞,在基因治疗及细胞治疗领域发挥作用。 The remarkable property of Intercellular transport by MPP - hPER1 of fusion Protein was observed in this Study.firstly a transcellular active peptides - PLNG was synthesized based on the analysis of the molecular composed of MPP family.Then, a fusion protein expression system was constructed by recombinant technique. Finally, the synthesized pep-tides - PLNG and MPP - hPER1 fusion protein were observed to have the property of transcellular membrane under different time, reaction temperature and cell line by the means of cell culture, transfection, and immunflurescence.It was suggested that this intercellular delivery by PLNG is time - independent, temperature - independent, energy - independent and efficient for a range of cell types.It is implied the PLNG can be applied to be a special biological macromolecule vector to delievery the functional protein, DNA and compounds through biological membrane to show the potential therapeutic application.
