摘要
目的 研究载脂蛋白 E(apolipoprotein E,apo E)基因多态性与早发冠心病 (coronary heartdisease,CHD)的相关关系及其对血脂水平的影响。方法 应用聚合酶链反应 -限制性片段长度多态性(polymerase chain reaction- restricted fragment length polymorphism,PCR- RFL P)基因分析方法 ,测定 5 2例早发 CHD、16 1例迟发 CHD患者和 180名对照者的 apo E基因型 ;血脂水平按常规方法测定。结果 发现的 5种 apo E基因型 ,分别为 E3/ 3、E4/ 4、E3/ 2、E4/ 3及 E4/ 2。早发 CHD组和迟发 CHD组 apo E4/ 3基因型和 ε4等位基因频率均高于对照组 (P<0 .0 1) ;进一步对两组 CHD患者的 apo E多态性进行分析 ,发现早发组ε4等位基因频率较迟发组为高 (P<0 .0 5 )。 apo E各等位基因型之间 ,TC和 L DL - C水平之间存在统计学差异 (P<0 .0 5 )。结论 apo E基因多态性与早发 CHD的发生发展有关并影响血脂的水平。
Objective: To detect the association of apolipoprotein E(apoE) gene polymorphism with early-onset coronary heart disease(CHD) and to evaluate the effect of apoE gene polymorphism on plasma lipid levels. Methods: The apoE genotypes of 52 early-onset CHD patients (aged 38-55 years), 161 aged CHD patients and 180 controls were determined by PCR-RFLP. Plasma lipid levels were measured by using routine methods. Results: Five genotypes of apoE, E3/3, E4/4, E3/2, E4/3 and E4/2 were detected in this study. Statistically differences in apo E4/3 and Ε4 frequencies between early-onset and aged CHD patients and controls were seen (P<0.05). The frequency of e4 in the early-onset CHD patients was higher than that in the aged CHD patients. Ε4 and Ε2 alleles also differently affected the plasma total cholesterol and low density lipoprotein cholesterol levels. Conclusion: These results suggest that apoE gene polymorphism is in association with the occurrence and development of early-onset CHD and it affects the plasma lipid levels.
