摘要
以滇Ⅰ型杂交粳稻高产组合榆杂 2 9保持系滇榆 1号B、恢复系南 2 9- 1和寻杂 36保持系滇寻 1号B、恢复系南 2 9- 2为材料。采用混合筛选法 ,从 2 0 0个RAPD引物中 ,筛出在两个组合保持系与恢复系间表现RAPD差异的 3个标记 :OPE - 1170 0 ,OPE - 1182 0 ,OPE - 11154 3 。分别随机提取 4个材料的 15 0个单株DNA ,构建 4个随机单株DNA群体。利用OPE - 1170 0 ,OPE - 1182 0 ,OPE - 11154 3 3个标记鉴别随机单株DNA群体。结果表明 :OPE- 1170 0 ,OPE - 1182 0 ,OPE - 11154 33个标记在 4个材料间均能扩增出明显的多态性产物 ,重复性好 ,4个材料的单株正确鉴别率分别为 :滇榆 1号B :97 3 % ,南 2 9- 1:96 % ,滇寻 1号B :95 3 % ,南 2 9- 2 :92 %。
The materials researched in the article are Dianyu I B,Nan29-1,Dianxun I B and NAN29-2, Dianyu I B is maintainer line of high yield combinationYuza29 and Nan29-1 is it's restoring line ,Dianxun I B is maintainer line of high yield combination Xunza29 and Nan29-2 is it's restoring line .Two combines are all belong to Dian I type Japonic Rice. The different marker OPE-11 700 ect between maintainer line and restoring line of two combinations was fond. We identified 600 random single plant DNA group with the marker OPE-11 700 ,OPE-11 820 ,OPE-11 1543 , which composed of 150 random single plant DNA of each material, the result shows the amplactive repeat of the markers are well and the single plant percent about four kinds of material identified as fellow:Dianyu I B : 97.3% ,Nan29-1: 96%,Dianyu I B:95.3%,Nan29-2: 92.0%.
