摘要
目的:建立α粒子照射诱发BEP2D细胞恶性转化模型。方法:α粒子照射永生化BEP2D细胞,通过细胞传代培养,最终经裸鼠成瘤实验,病理切片鉴定,瘤组织培养获取肿瘤细胞系,角蛋白多克隆抗体免疫组化鉴定细胞上皮来源。结果:(1)α粒子照射BEP2D细胞后,培养35代,54代细胞接种裸鼠成瘤,照后20代BEP2D细胞及未照射永生化BEP2D细胞接种裸鼠不成瘤;(2)瘤体病理切片鉴定为鳞状上皮癌;(3)瘤组织培养得到肿瘤细胞系2个,角蛋白多抗免疫组化实验,细胞胞浆强阳性,证实为上皮来源。结论:1.5Gyα粒子照射诱发BEP2D细胞恶性转化,成功建立α粒子照射诱发肺癌的细胞研究模型。
Objective: To develop a malignant transformation cell model induced by α particles for lung cancer research. Methods: This procedure involves cell culture, irradiation, tumorigenicity, and establishment of tumor cell line, keratin staining. Results: (1) Upon inoculation into nude mice, cells produced tumors; (2) Tumour isolated from nude mice was characterized as squamous epithelial cancer;(3) Keratin staining was positive, tumor cell lines RHT35 and RHT54 were set up. Conclusion: Malignant transformation of BEP2D cell lines were produced after irradiation of 1.5 Gy α particles, cell model induced by α particles for lung cancer research was set up.
