摘要
目的 :以荧光标记的mRNA差异显示技术分离食管癌及其正常食管粘膜组织相关的差异基因片段。方法 :结合GenomyxLRTM DNA测序 /差异显示系统及GenomyxSCTM 荧光图象扫描系统 ,以及配套的FluoroDD和HI EROGLYPHmRNAProfileKit,采用荧光标记的mRNA差异显示技术进行检测。结果 :从人食管癌及正常食管粘膜组织间 ,同时直接分离出了相关的癌基因或抑癌基因片段 74条。结论 :该技术方便、快捷 。
Aim: To isolate associated gene fragment in human esophageal cancer and normal mucosa tissues by using nonradioactive isotope fluorescence label mRNA differential display technique. Methods: The associated gene fragments were detected by using fluorescence label mRNA differential display technique with Genomyx LR TM DNA Sequence /Differential Display system, Genomyx SC TM Fluorescent Imaging Scanner system , as well as FluoroDD and HIEROGLYPH mRNA Profile Kit . Results: Seventy four associated cDNA fragments were isolated from esophageal cancer and normal mucosa tissues. Conclusions: This technique is convenient and prompt, and is worth popularizing and applying.
