食管粘膜不典型增生组织中端粒酶活性的定量及定性检测 被引量:5

摘要 目的 :探讨食管粘膜不典型增生组织中端粒酶活性的表达。方法 :采用聚合酶链反应 酶联免疫吸附测定法 (TRAP ELISA)定量及TRAP 银染法定性技术 ,对 15例食管不典型增生组织及 12例正常食管粘膜进行端粒酶活性检测。结果 :15例不典型增生组织中 ,9例端粒酶活性呈阳性 (其中 8例轻、中度不典型增生中 4例呈阳性 ;7例重度不典型增生中 5例呈阳性 ) ,12例正常食管粘膜中 ,仅 1例端粒酶活性呈阳性。端粒酶活性平均A值在轻、中度不典型增生与重度不典型增生组织中分别为 0 .945± 0 .6 0 1和 1.2 5 6± 0 .785 ,正常食管粘膜组织为 0 .376± 0 .2 11。正常食管粘膜与不典型增生组相比 ,其端粒酶活性检出率和平均A值组间差异均存在显著性 (P <0 0 1)。结论 :端粒酶的激活在食管癌的发生与发展中起着重要作用 ,其活性检测可望成为监测食管癌发生及早期诊断的良好指标。 Aim: To investigate the expression of telomerase activity in dysplasia tissues of the esophagus. Methods: Telomerase activity in specimens from 15 cases with dysplasia tissues and 12 cases of normal esophageal mucosa was measured quantitatively by the TRAP ELISA and qualitatively by TRAP silver staining. Results: It was found that telomerase activity was positive in 9 of the 15 dysplasia tissues but only in 1 of the 12 normal esophageal mucosa. The mean level of telomerase activity in high dysplasia, middle and mild dysplasia, and normal esophageal mucosa were 1.256±0.785,0.945±0.601 and 0.376±0.211, respectively. There were significant differences in telomerase positive expression rate and the mean level of telomerase activity between dysplasia tissue and normal esophageal tissue( P <0.01). Conclusion: The activation of telomerase may play an important role during carcinogenesis and progression of esophageal cancer. Telomerase activity might be a good marker in estimating the occurrence, development and early diagnosis of esophageal cancer.
