摘要
目的 利用人类幽门螺杆菌急性感染的小鼠模型研究Hp对胃壁细胞及H + K +ATP酶mRNA基因表达的影响。方法 2 0只SPF级BALB/C小鼠分为两组 ,实验组和对照组 ,实验组动物每只灌胃 0 .4mlHp菌液 ,连续 5d ;而对照组予生理盐水灌胃。 2周后处死动物 ,取胃黏膜分别用于尿素酶试验、Giemsa染色 ;电镜观察壁细胞形态 ,用图象分析软件分别计算出两组分泌小管面积分数 ;用RT PCR测定H+ K+ ATP酶mRNA基因表达的情况。结果 实验组动物胃黏膜尿素酶试验及Giemsa染色均为阳性 ;图象分析软件结果显示分泌小管面积分数 ,试验组为 1.96± 0 .0 8/ 10 4 ,对照组为 4.2 9± 2 .30 / 10 4 ,两组比较有显著性差异 ;用RT PCR测定H+ K+ ATP酶mRNA基因表达结果显示实验组为 0 .92± 0 .14,对照组为 1.0 4± 0 .14,两组比较有显著性差异。结论 幽门螺杆菌急性感染后 ,胃酸分泌明显减少 ,减少的原因考虑与下调H+ K+
Objective The effect of Hp on gastric acid secretion has not come to a conclusion. Many research studies were conducted in vitro. The aim of this study was to find out the changes of gastric parietal cell and H + K +ATPase mRNA expression after acute H pylori (SS1) infection in mice. Methods Twenty 7 week old SPF BALB/C mice were divided into two groups, experimental group and control group according to the body weight, each group ten mice with five males and females each. Mice in the experimental group were inoculated orally H pylori liquid (Sydney Strain 1, SS1) 0.4 ml (10 9CFU) per mouse every day for five days while those in the control group were given placebo of equivalent quantity. Two weeks after the last bacteria inoculum, all mice were sacrificed. Hp colonization were detected by HE stain ,Giemsa stain and urease test. Ultrastructures of parietal cells were observed by electromicroscope. Gastric H + K +ATPase mRNA expression was measured by RT PCR. Results The experimental group were 100% infected by Hp. RUT and Giemsa stain were all positive. HE stain showed an acute inflammation with neutrophilic granulocyte infiltration. Whereas RUT and Giemsa stain of the control group were all negative , and no abnormality was seen by HE staining. In the experimental group, the average ratio of AC to AT (AC mean area of secretory canal, AT area of the parietal cell) was 1.96±0.08/10 4 , significantly less than that (4.29±2.30/10 4) of the control group ( P <0.01). The average H + K +ATPase/β actin mRNA RT PCR ratio of the experimental group was 1.048± 0.055, significantly lower than that (1.223±0.0073) of the control group( P <0.01). Conclusion These data suggest gastric acid secretion is inhibited after the acute H pylori infection, and one of the mechanisms by which Hp inhibits parietal cell acid secretion is down regulation of H + K +ATPase mRNA expression .
