摘要
利用PE270A_HT型高效毛细管电泳仪 ,采用无胶筛分电泳 ,以羟丙基甲基纤维素 (HPMC)为筛分介质 ,对三裂叶豚草 (AmbrosiatrifidaL.)的聚合酶链反应 (PCR)产物进行了分离分析 ,显示了高效、快速、无污染的毛细管电泳技术在植物DNA片段多态性分析方面的巨大的潜力。
Capillary non _ gel sieving(NGS) electrophoresis was used for the direct analysis of polymerase chain reaction(PCR) products of Ambrosia trifida L. with HPMC(hydroxypropyl methyl cellulose) as a sieving medium for NGS. High performance capillary electrophoresis has a high repeatability with RSD of 0.25%(n=5) for retention time of peak in the separation of DNA fragment(0~2 000 base pair)produced by PCR. It is found that high performance capillary electrophoresis has great potentialities in the analysis of polymorphic species of DNA fragment.
