肝细胞膜子孢子抗原受体的分离及特性的初探——β型抗独特型单克隆抗体亲和层析法

摘要 本文通过标记抗原的活体分布试验证明,抗独特型单克隆抗体D3作为抗原的内影像对肝细胞有一定的亲和性,以D3探针制备的D3-Sepharose 4B亲和柱自大鼠肝细胞膜蛋白中分离纯化出分子量为31Kd的子孢子抗原受体。该受体蛋白能封闭子孢子入侵肝细胞,使其失去对宿主的感染力。提示抗原受体在抗虫免疫中有着不可忽视的作用,有理论和应用研究的价值,值得进一步探讨。 D3, as an internal image of sporozoite antigen, had affinity to hepato-cytes, which was observed by 125I-D3 distribution test in vivo, and as probe of anti-genicrecrptor expressed on hepatocytes,could be used to isolate sporozoite-specif ic protein with the molecule weight of 3lKd from soluble membrane protein of rat hepatocytes. Incubation of sporozoites in vitro with affinity-purified membrane protein of hepatocytes abolished their infectivity, which indicated that this protein could function as receptor or mediator for the sporozoites in the process of invasion of the hepatocytes
