摘要
B_3为我室建立的带有HBsAg表位内影像的单克隆抗独特型抗体(mAb_2)。本文采用脾脏内和尾静脉内注射途径,用B_3免疫BALB/c鼠,获得了具有抗-HBs结合活性的同系单克隆抗-抗独特型抗体(mAb_3)3B_8株。这一结论基于以下两点观察:(1)3B_8能与纯化的HBsAg结合。纯化的HBsAg或抗-HBs均能以剂量依赖形式抑制这一结合;(2)3B_8能以剂量依赖形式抑制抗-HBs与B_3的结合。小鼠在从未接触HBsAg的情况下,单独用B_3免疫,获得了具有抗-HBs结合活性的单克隆抗-抗独特型抗体3B_8,进一步证明我室过去建立的mAb_2B_3株,确实带有HBsAg表位内影像。
Previously, we generated a hybridoma (B3) which secrets monoclonal anti-Id (mAb2) antibody that bears the internal image of HBsAg epitope. To characterize the mAb2 further, they were used to immunize BALB/c mice by intrasplenic and intravenous injection. Syngeneic monoclonal anti-anti-Id (3B8, mAb3) which bear the binding activity of anti-HBs were derived. This conclusion was based on the following observation: (l)the mAb3 can bind to purified HBsAg and this binding can be blocked by HBsAg or anti-HBs(Abl)in a dose-dependent manner; (2) the mAb3 can block the binding of anti-HBs (Abl) to mAb2(B3)in a dose-dependent mannerj and (3) the immunized mice with mAb2 (B3) were not exposured to HBsAg.
In conclusion, the production of mAb3 that bear the anti-HBs binding activity in mice in the absence of any exposure to HBsAg indicates that the mAb2(B3) may indeed carry the internal image of HBsAg epitope. Such anti-Id (Ab2) may be used as suro-gate antigen for the induction of anti-HBs response.
