RT-PCR测定乳癌及腋窝淋巴结微量组织MUC1及GST-π基因的表达 被引量:2

摘要 目的 研究 MUC1及 GST-π m RNA在乳癌腋窝清扫淋巴结中表达与乳癌发生、发展、预后转归及临床耐药的相关性。方法 用 RT- PCR技术检测 2 0例乳腺癌病人的 1 0 8个 HE染色阴性淋巴结 MUC1及 GST-π m RNA的表达。结果  MUC1及 GST-π m RNA在 HE染色阴性淋巴结中阳性检出率分别为 66%及 39% ,在乳癌组织、HE阳性淋巴结及正常乳腺组织中阳性率分别为 1 0 0 % /88%、1 0 0 % /71 %及 1 0 0 % /0。结论 HE阴性淋巴结中出现 MUC1阳性表达表明存在肿瘤微转移灶 ,MUC1 m RNA检测可提高淋巴结微转移诊断率 ;GST- π在肿瘤启动和促进阶段可表达 ,是一种重要的乳腺癌生物学标志物 ;乳癌组织中耐药性普遍存在 ;MUC1及 GST- π m R-NA在腋淋巴结的表达可能是预后不良的标志 ,二者联合检测对乳癌早期诊断及预后判断有重要意义。 Objective To study MUC1 and GST π mRNA expression in axillary lymph nodes and its clinical significance.Methods Expression of both MUC1 mRNA and GST π mRNA was detected by RT PCR in 108 histologically negative lymph nodes obtained from 20 breast cancer patients.Results Of 108 lymph nodes,66%/39% were found to express MUC1 mRNA/GST π mRNA.And the positive rates of MUC1 and GST π mRNA in cancer tissues,HE positive lymph nodes and normal breast tissue are 100%/87%,100%/80%,100%/0,respectively.Conclusion Expression of MUC1 mRNA in histologically negative lymph nodes suggests the presence of breast cancer micrometastases;GST π ,whose expression is detected as cancer occurring and developing,can be used as a breast tumor marker;Drug resistance exists in almost all the breast cancer,The expression of MUC1 mRNA and GST π mRNA in lymphnodes may be a indication of poor prognosis.The detection of all these two genes together by RT PCR has higher sensitivity and significance.
