神经肽Y5受体反义脱氧寡核苷酸的减重降脂作用 被引量:3

摘要 目的 : 探计神经肽 Y5反义脱氧寡核苷酸对高营养饮食诱导的肥胖大鼠模型的拮抗作用。方法 : 建立高营养饮食诱导的肥胖大鼠模型 ,行侧脑室内插管 ,观察肥胖大鼠注射神经肽 Y5受体反义脱氧寡核苷酸、错义脱氧寡核苷酸及生理盐水后体重和进食量的变化。结果 : 注射神经肽 Y5受体反义脱氧寡核苷酸的肥胖大鼠 ,体重和进食量明显减少 ,与注射前比较有显著性差异 ( P<0 .0 1 ) ;注射错义脱氧寡核苷酸和生理盐水对照组这两项指标未见明显下降 ( P>0 .0 5 )。结论 : 侧脑室注射神经肽 Y5受体反义寡核苷酸对营养性肥胖大鼠有明显的抑制进食和减轻体重的效应。 Objective:To investigate the inhibitory effects of intracerebroventricular injection of neuropeptide Y Y5 receptor antisense and mismatch oligodeoxynucleotide and sodium chloride on food intake and body weight in diet induced obese rats.. Methods:Male SD rats were fed a standard diet(n=7)and high nutrition diet(n=21)for 7 weeks. Then the rats received cannula implantation into the right lateral ventricle. After 7 days of recovery, intracerebroventricular injections of neuropeptide Y Y5 receptor antisense and mismatch oligodeoxynucleotide 10μl(5μg/μl)and 10μl saline were given three times a day for 2 days.The changes of body weight and food intake were measured. Results:In diet induced obese rats, the inhibition of body weight and food intake were observed after intracerebroventricular neuropeptide Y Y5 receptor antisense oligodeoxynucleotide injection at the first day(P<0.01).But in mismatch oligodeoxynucleotide and sodium chloride injection group, the effects were not significant(P>0.05). Conclusion:Neuropeptide Y Y5 receptor antisense oligodeoxynucleotide can inhibit the food intake and reduce the body weight in diet induced obese rats after intracerebroventricular injection.
