摘要 目的 :建立一种猪冠状动脉平滑肌细胞急性酶分离法并在分离的细胞上研究其离子通道特性。方法 :采用木瓜蛋白酶 ,胶原酶和透明质酸酶分离猪冠状动脉平滑肌细胞 ,用膜片钳单通道记录法记录通道电流。结果 :该方法分离过程简单 ,所获细胞数量多 ,形态正常 ,在这些细胞上容易记录到正常的KCa电流。结论 :应用本方法易得到单个活性正常的 。 Objective: To find a new technique for isolation of single porcine coronary artery smooth muscle cells and to characterize the calcium-activated potassium channels(K Ca ) in these cells .Method:Single artery smooth muscle cells were isolated by employing papain collagenase and hyaluronidase enzyme dispersion medium.Single-channle currents were recorded with patch-clamp technique. Result: The isolation method can produce large number of single artery smooth muscle cells in normal form and the K Ca currents can be easily recorded. Conclusion:The enzymatical dissociation method was proved simple ,which provided a useful extension for understanding the physiology and biology in the study of ionic channels.
