增强细胞渗透性对酿酒酵母ATP合成活力的作用研究 被引量:3

摘要 研究了一株啤酒酵母在不同培养条件下生长的细胞渗透性及其ATP合成活性 实验检测了氧限制条件下生长酵母的细胞渗透性 ,并选用有机溶剂、超声波和气流干燥等通透方法 ,对培养细胞的渗透性进行增强处理 ,考察细胞的渗透性变化对其ATP生产活性的作用 结果表明 :渗透性较差的细胞 ,其ATP生产活力也相对较低 催化腺苷 (Ado)生产ATP转化率低于 2 0 %的培养细胞 (A -细胞 )经通透处理后 ,转化率能提高 30 %~ 10 0 % ;渗透性较好、转化率较高的培养细胞 (M -细胞 )经通透处理后 ,其完成ATP合成反应的时间相对能缩短 1~ 2h ,催化反应速度明显提高 经通透化处理的细胞能更有效地释放胞内物质和表现出较高的乙醇脱氢酶 (ADH)和己糖激酶 (HK)活性 。 This correlation between cell permeability and the cell enzymatic activity for ATP production was studied in the brewer's yeast,sacchromyces cerevisiae TB6.The cells (A-cells) cultured aerobically have less permeability and activity than the cells (M-cells) cultured oxygen-limited.When these cells were treated with 2% (v/v) toluene,ultrasonic disruption and air drying respectively,the conversion rate into ATP increased from 20% to 30%~100% in A-cells,and the reaction time for ATP production decreased from 4~5 to 2~3 hours in M-cells.The permeabilized cells showed better biocatalysed activity for ATP production.Our results indicate that the permeabilization of yeast cells is related to its culture conditions,and it may reflect partly the cellular enzyme activity for ATP production.
