端粒酶逆转录酶在原发性结肠癌中的表达

Expression of Human Telomerase Reverse Transcriptase in Primary Colorectal Carcinoma

目的 探讨原发性结肠癌组织中端粒酶逆转录酶 (humantelomerasereversetranscriptase ,hTRT)的表达和端粒酶活性的关系以及在结肠癌分子生物学诊断的临床意义。方法 用免疫组化和TRAP法分别观察 5 4例原发性结肠癌组织和正常结肠组织中hTRT和端粒酶的活性 ,比较分析它们的相关性。结果 hTRT阳性率83% (4 5 / 5 4) ,端粒酶活性阳性率 85 % (4 6 / 5 4) ,在结肠癌组中 3例术前化疗病人hTRT表达呈 2例弱阳性 1例阴性 ,结肠癌淋巴结转移的hTRT阳性率达 92 % ,10例正常结肠组织均为阴性。结论 hTRT在结肠癌中具有癌细胞的特征性 ,并与端粒酶活性呈正相关。hTRT可作为结肠癌诊断的敏感的指标 。

Objective To investigate the expression of human telomerase reverse transcriptase (hTRT) in primary colorectal cancer and the relationship between the telomerase activity and the clinical significance of the colorectal cancer molecular diagnosis.Methods 54 cases of primary colorectal carcinoma and 10 normal colon tissues were analyzed for hTRT by immuno histochemistry and meanwhile the telomerase activity was measured by TRAP assay.Results The TRAP assay showed there were 85% (46/54) samples which presented a telomerase activity, and the expression rate of hTRT was 83%(45/54), in which , 37/40 patients with lymph node metastases were 92%; 3 patients who accepted chemotherapy before surgical operation showed a weak expression in two samples and one was negative. 10 normals were all negative.Conclusion hTRT, as an index of colorectal cancer, shows a good application in colorectal cancer diagnosis, and the immuno histochemistry of hTRT is easier than TRAP assay in testing telomerase activity in colorectal cancers.
