摘要
目的 :探讨不同细胞因子对大肠癌细胞 Fas配体 (Fas L )表达水平的调控作用。方法 :采用 Western印迹法测定了两种细胞因子 (1× 10 6 U / L TNF-α或 IFN-γ)处理后大肠癌细胞 Fas L表达水平的变化。结果 :所检测的 6株大肠癌细胞全部表达 Fas L蛋白 ;TNF-α和 IFN-γ处理 DL D- 1细胞 10 h,显著上调其 Fas L蛋白的表达水平 ;TNF-α和 IFN-γ以及 PMA +离子霉素 (ionomycin)处理 SW6 2 0细胞 10 h,也显著上调 SW6 2 0细胞的 Fas L蛋白表达水平。 TNF-α和 PMA +离子霉素处理SW6 2 0细胞 48h进一步增加 SW6 2 0的 Fas L 蛋白表达 ,IFN- γ处理 SM6 2 0则表达增加不明显。结论 :TNF- α和 IFN- γ等细胞因子具有上调某些大肠癌细胞 Fas L
Objective: To investigate the role of TNF-α and IFN-γ in modulation of Fas ligand(FasL) expression in human colon carcinoma cells. Methods: A total of 6 human colon cancer cell lines were examined for FasL protein expression and for TNF-α and IFN-γ modulation of FasL expression by Western blot method. Results: The results showed that FasL protein was expressed in all of these 6 cancer cell lines.FasL expression was up-regulated by TNF-α and IFN-γ for 10 h in DLD-1 colon cancer cells; FasL expression was also up-regulated by TNF-α and IFN-γ as well as PMA plus ionomycin for 10 h in SW620 cells, and further up-regulation was got by TNF-α and PMA plus ionomycin for 48 h in SW620 cells,whereas slight up-regulation was obtained by IFN-γ. Conclusion: These data suggest that TNF-α and IFN-γ may enhance FasL expression in tumor cells and may facilitate to escape from the immune surveillance of the host.
