增殖细胞核抗原表达对激光诱导血管平滑肌细胞凋亡的影响

摘要 目的探讨体外培养的血管平滑肌细胞(VSMC)增殖细胞核抗原(PCNA)表达对其凋亡的激光诱导率的影响。方法组织贴块法培养VSMC,饥饿法同步化,在不同的刺激因子作用之后,免疫组化法检测细胞PCNA表达率,各组细胞经510.6nm的铜蒸汽激光照射,TUNEL法记数细胞凋亡率。结果PCNA表达高的细胞,在激光的诱导下易发生凋亡;相反,PCNA表达低细胞,凋亡诱导率较低。结论经不同生长因子刺激后VSMC的PCNA表达率不同,PCNA表达率的不同对激光的凋亡诱导率产生影响。 Objective:to investigate the effect of vascular smooth muscle cells proliferative activity and cell cycle on apoptosis rate induced by Copper Vapor laser irradiation.Methods:VSMCs were synchronized and divided into3groups:①10%NCS group;②PDGF group;③40%FBS group.Then different growth factors were added into each group.PCNA immunohistochemical staining was used to measure SMC proliferative status,at the same time,apoptosis rate of each group was analyzed by TUNNEL assay after irradiated with Cooper Vapor laser.Results:positive rate of PCNA of group②and③is higher than group①,and the apoptostic rate had the same tendency.Conclusion:cells at the proliferative stage are easy to be apoptosized under the in-duction of laser,while the apoptosis rate of relatively stable cells is low.
