摘要
目的 探讨新生猪缺氧时循环内皮细胞的形态学变化。方法 复制新生猪缺氧模型 (HM) ;从富含血小板血浆中分离循环内皮细胞 (circulatingendothelialcell,CEC) ;采用ⅧR :Ag间接免疫荧光进行检测 ,黄绿色荧光着色细胞为循环内皮细胞。结果 对照组CEC为 2 .17± 0 .82× 10 9/L ,HM组为 3.0 7± 0 .93× 10 9/L ,两组有显著差异 (P <0 .0 1) ;在形态学上CEC分为 3种 :①核清晰可见 ,胞浆丰富 ;②核隐约可见 ,胞浆少见 ;③核缺失 ,细胞明显皱缩。Ⅷ :Ag荧光检测以上 3种细胞均发现较多的黄绿色荧光着色。结论 HM组中CEC明显增多 ,提示缺氧对血管内皮细胞有明显损伤作用 ,CEC可作为血管损伤的指示物之一。
Objective To detect the change of the circulating endothelial cells(CEC) in the hypoxic damage. Methods To make the models of the hypoxic damage(HM);Isolating CEC from platelet rich plasma by Hladovec's method;Ⅷ:Ag indirect immune fluorescence method to identify CEC. Results The CEC of the normal contrast group was 2.17±0.82×10 9/L and HM was 3.07±0.93×10 9/L.The CEC of two groups had much different ( P <0.01).The CEC in HM could be divided into three kinds cells ①The CEC was clear nucleus and rich plasma;②The CEC was nucleus indistinctly visible and seldom plasma that were compared with other CEC and ③ a nucleus,crumple,fold shape.There were a lot of yellow green fluorescent substances in the CEC by Ⅷ:Ag indirect fluorescent test. Conclusion There were much more CEC in the HM than those in contrast grope.It has hypoxic effect in model damaging the endothelial vascular cells,the CEC can be one of indicated substances of the vascular damage.
