摘要 目的:建立一种有效、快速的HPLC法测定血液中微量的甘草酸,并对其药动学进行初步研究。方法:从甘草中提取、纯化甘草酸单钾盐,给小鼠静脉注射和灌胃,定时取出血样,使用HPLC测定其血药浓度,数据使用3P87程序处理,绘制药时曲线。结果:小鼠灌胃(500mg/kg)2.2%甘草酸单钾盐,药时曲线符合双室模型,α相t_(1/2)=15.97min,β相t_(1/2)=200.05min,AUC=2453.96(μg/ml)·min,Cl(s)=3.642μg/min(μg/ml)。小鼠静脉注射0.1%的甘草酸单钾盐40mg/kg药时曲线符合双室模型,α相t_(1/2)=5.128min,β相t_(1/2)=51.196min,AUC=7477.642(μg/ml)·min,Cl(s)=0.0956μg/min(μg/ml)。结论:HPLC法是一种方便,可靠,快速的检测甘草酸单钾盐血药浓度的方法。 To find a practive and quick method determining GK concentration in blood by HPLC and to study its pharmacokinetics. GK from Glycyrrhiza uralensis Fisch was extracted and purified. GK were administrated to mice by iv. and ig. The Concentration of GK in blood at expected time was determined by HPLC. The data was dealed with by 3 P 87 programe. The concentration - time curve of iv 0.1% GK 40 mg/kg is in accordance with tow-compartment model. t1/2α= 5.128 min,t1/2β= 51.196 min,AUC = 7477.642 (μg/ml) ·min,Cl(s) =0.0956 μg/min(μg/ml). The c- t curve of ig. 2.2% GK 500 mg/kg is in accordance with tow compartment model, t1/2α=15.97 min,t1/2β= 200.05 min,AUC = 2453.96(μg/ml)·min,Cl(s) = 3.642 μg/min(μg/ml). HPLC is a quick and practive method to determin the concentration of GK in blood.
