摘要
目的 观察脑缺血对 IL - 1RI的表达及电针对其表达的调节。 方法 采用 SD大鼠大脑中动脉线栓法制备 MCAO缺血模型 ,应用原位杂交和免疫组化的方法观察脑缺血及电针对大鼠大脑皮层白细胞介素 - 1受体 ( 型 ) (interleukin- 1receptor type ,IL - 1RI) m RNA和蛋白表达的影响。 结果 在正常及假手术大鼠 IL - 1RIm RNA和蛋白阳性细胞在皮层呈基础水平表达 ;脑缺血再灌注后 2 h IL -1RIm RNA阳性细胞表达增加 ,6 h达高峰。脑缺血后 IL - 1RI蛋白在缺血侧皮层表达明显增加 ,再灌注后 12 h达高峰。电针可显著降低 MCAO大鼠大脑皮层 IL - 1RIm RNA和蛋白的表达。 结论 脑缺血后 IL - 1RIm RNA和蛋白表达增加 ,电针可降低 IL - 1RIm RNA和蛋白的表达 ,从而减轻缺血后 IL -
Objective To investigated the effects of electroacupuncture (EA) on the expression of Interleukin 1 receptor type Ⅰ(IL 1RI)mRNA and protein in focal middle cerebral artery occlusion rat. Methods In situ hybridization and immunohistology was used to detect the expression of IL 1RImRNA and prortein in the brain of middle cerebral artery occlusion rats. Results The normal and sham rats showed a basic expression of IL 1RImRNA and protein in cortex. The cells express IL 1RImRNA were increased after 2 h after reperfusion, maximally increased in ischemic cortex by 6 h of reperfusion and prolonged up to 24 h. The number of IL 1RI protein positive cells increased after ischemia. It increased by 6 h and reached the peak at 12 h in ischemic cortex after reperfusion. EA could downregulate the expression of IL 1RImRNA and protein in ischemic cortex. Conclusion The results indicated that the downregulated expression of IL 1RImRNA and protein may be one of the mechanisms of EA protect ischemic damage.
