摘要 建立人血浆中麦考酚酸酯活性代谢物麦考酚酸的 HPL C测定法。以 10 %三氯乙酸的乙醇溶液直接沉淀血浆蛋白 ,色谱柱为 L ichrom sorb RP- 18(2 0 0× 3.9mm ,5μm) ,预柱为岛津 ODS(5× 3.9m m,10μm ) ,流动相为乙腈 -0 .0 5 %磷酸溶液 (4 0∶ 6 0 ) ,检测波长 2 5 0 nm ,以尼群地平为内标。麦考酚酸的保留时间为 6 .85 min,血浆定量线性范围 0 .2 36~ 2 3.6 0μg/m l,方法回收率为 99%~ 10 5 % ,日内、日间 RSD小于 7%。本法简便快速 。 A HPLC method for the rapid determination of mycophenolic acid, bioactive metabolite of mycophenolate mofetil, in human plasma had been developed. The plasma protein was precipitated by the addition of 10% trichloroacetic acid solution in ethanol. Lichromsorb RP 18 (200×3.9 mm, 5 μm) column was used with a guard column of Shimadzu ODS(5×3.9 mm,10 μm), using a mixture of acetonitrile 0.05% phosphoric acid solution (40∶60) as the mobile phase. The eluate was detected at UV 250 nm and nitrendipine as the internal standard. The retention time of mycophenolic acid was about 6.85 min and a good linearity was shown over the concentration range of 0.236~23.60 μg/ml, the recoveries were 99%~105%, and the intra day or inter day precisions were less than 7%. This HPLC method is simple, rapid and accurate.
