大鼠淋巴树状突细胞与巨噬细胞的形态学观察

摘要 对大鼠淋巴树状突细胞(LDCs)和巨噬细胞(Mφs)的观察结果如下:相差镜下 LDCs 突起持续伸缩而 Mφs 呈静止球形;扫描电镜下 LDCs 表面光滑而 Mφs 表面多皱褶和微绒毛;透射电镜下 LDcs 细胞器很少而 Mφs 细胞器发达;免疫荧光梁 Ia 抗原,LDCs 呈强阳性反应而 Mφs 弱阳性或阴性;S-100蛋白免疫染色,LDCs 呈强阳性反应而Mφs 阴性;ACP 反应 Mφs 强阳性而 LDCs 弱阳性或阴性。因此 LDCs 和 Mφs 可用 S-100蛋白和 ACP 两种染色来判断区分。在悬液细胞,相差镜和扫描电镜检查也有鉴别意义。 The results of our observations are as followes:(1)By phase contrast microscopy,the lymphoid dendritic cells(LDCs)could be easily distinguished from macrophages(Mφs) by their consistently moving cytoplasmic processes,and Mφs were found to be spheri- cal and stationary.(2)By scanning EM,the LDCs exhibited a smooth cell surface, while Mφs show a ruffled surface,microvilli and cristae.(3)Transmission EM revea- led that LDCs lacked organelles while Mφs were rich in them.(4)After immumoflu- orescence staining,LDCs were found to have a strongly positive reaction,while Mφs, have a weakly Positive or negative reaction,(5)After S-100 protein staining,LDCs showed a strongly positive reaction,while Mφs had a clearly negative one.Diserimi- nation of LDCs and Mφs may be performed using ACP staining in addition to S-100 protein staining.Phase contrast microscopy and scanning EM might be helpful in thier differentiation.
