摘要
探讨视网膜脱离(RD)后色素上皮(RPE)细胞的凋亡。方法 制造有色眼家兔的RD模型,以流式细胞仪(FCM)技术分析RD后28天内RPE细胞的凋亡变化。结果 RD后8h RPE细胞凋亡显著增加,第7天达顶峰,凋亡细胞平均数由正常的131.67±35.67上升至1196.2±238,凋亡率由正常的0.70%±0.21%上升至5.98%±1.21%,然后缓慢下降,第28天仍显著高于正常。结论 RD可导致RPE细胞的凋亡。RD后28天内,随时间延长,RPE凋亡增加。
Objective To analyze the apoptosis of retinal pigment epithelial (RPE) cells after retinal detachment(RD). Methods The experimental RD models in pigmented rabbits were established,and the apoptosis of RPE cells was assessed by low cytometry and observed with the microscope at the 8 hour, 1st,4th,7th, 14th and 28th day after RD. Results The apopto- sis of RPE cells began to increase at 8 hr after RD and reached peak on the 7th day after RD,the average of apoptotic cells from 131. 67 ± 35. 67 in the normal group increased to 1196. 2 ± 238 in RD group, the apoptotic rate from 0. 70% ± 21 % in the normal group increased to 5. 98% ± 1. 21 % after RD. The apoptotic cells and the apoptotic rate were much higher in RD group than those in the normal group at the 28th day. Conclusion The apoptosis of RPE cells might be caused by RD. With the RD time last- ing ,the RPE apoptosis increased in 28d after RD. FCM could he used to assess the apoptosis of RPE cells accurately and rapidly.
