球后注射8-氯腺苷后眼内药代动力学研究 被引量:2

摘要 观察新型抗代谢药物8-氯腺苷球后注射后眼内的分布情况,以探讨该药物用于视网膜和脉络膜新生血管性病变治疗的可行性。方法 采用高效液相色谱分析法分别测定单次球后注射及反复多次球后注射8-氯腺苷后,眼内的药物分布情况。结果 单次球后注射8-氯腺苷后,眼后段的各组织中均检测到不同浓度的8-氯腺苷以及其代谢产物8-氯腺瞟呤。反复多次球后注射8-氯腺苷后未见蓄积效应。结论 球后注射8-Cl-A后,该药在眼后段具有良好的分布。多次应用眼内无明显的蓄积作用。 Objective To elucidate the intraocular distribution of anti-metabolic agent 8-chloroadenosine following retro- bulbar injection and its potential role in the clinical treatment of retinal and choroidal neovascularization. Methods The intrao- cular distribution of 8-chloroadenosine following single and repeated retrobulbar injections was studied by high performance liquid chromatography, respectively. Results 8 - chloroadenosine and its metabolite 8 -chloroadnine were found in tissues and ocular samples from the posterior segment following single retrobulbar injection. No cumulative effect of 8-chloroadenosine was observed after repeated retrobulbar injections. Conclusion 8-chloroadenosine can be well distributed in the posterior segment following retrobulbar injection without cumulative effect.
