肝细胞脾脏内移植对实验性暴发性肝功能衰竭疗效探讨

EFFICACY OF INTRASPLENIC TRANSPLANTATION OF FRESH HEPATOCYTE IN TREATMENT OF EXPERIMENTAL FULMINANT HEPATIC FAILURE IN RATS

本文探讨了同种异体新鲜活性肝细胞移植对 CCl_4诱导之暴发性肝功能衰竭(Fulminant Hepatic Failure,FHF)动物的治疗效果。移植两周后,移植组动物存活率明显高于对照组。动物模型制备前、后,各组动物血生化平均值具有显著性差别;CCl_4中毒后肝组织学表现为细胞大片坏死、脂肪变性及空泡变性,提示肝脏功能、结构受到严重损害。而存活率的改善证实了肝细胞脾脏内移植能够对 FHF 动物提供足够的代谢支持作用。

Fifty-eight Wister rats with experimental fulminant hepatic failure (FHF) were randomly divided into th ree groups:group Ⅰ:transplantation group;group Ⅱ:controls; group Ⅲhas the same treatment as group Ⅰ,but is used for histologic examination. The survival rate of G Ⅰ and G Ⅱ two weeks after CCl4 administration was 50% and 20% respectively.The difference in survival between G Ⅰ and G Ⅱ was statistically significant,P<0.05.It proved that intrasplenic hepatocyte transplantation elevated the survival rate of FHF rats.Significant elevations(P<0.0005) were also observed for SGPT,SGOT,AKP and TB values,amounting to fivefold after administration of CCl4.Histologic examination showed that liver tissues after CCl4 administration presented major necrosis,fatty degeneration and vacuolization of hepatoeytes.It indi- cates that the function and structure of liver are seriously impaired.We conclude that intrasplenic hepatocyte transplantation supplies a good metabolic support for the rat with FHF.
