摘要
本文运用单因素试验设计及正交试验设计理论确定高产L-乳酸突变菌株NAF-032的最优种子培养基及培养条件。最优种子培养基(g/1):葡萄糖80.0,氯化铵2.0,NaH2PO4 0.3,KH2PO4 0.3MgSO4·7H2O 0.25,ZnSO4·7H2O 0.05。最优种子培养条件:温度34℃,转速 200r/min,装液量60ml(250ml三角瓶),一次性添加CaCO310.0g/1用于调节pH值,种子培养时间22h。
The optimal preculture medium and culture conditions for L-lactic acid high-producing strain Rhizopus oryzae NaF-032 were developed by single factor and orthogonal design theory. From the shake-flask experiments, the optimal preculture medium(g/l) and culture conditions were: glucose 80.0, NH4 Cl 2.0, KH2P O4 0.3, NaH2PO4 0.3, MgSO4·7H2O 0.25, ZnSO4·7H2O 0.05;34,200r/min, loading 60 ml in 250ml flask, 10.0g/L CaCO3 was added to control the pH of both, 22h culture age.
