摘要
用玻璃微针徒手切割小鼠早期胚胎,分割的裸半胚在体外培养条件下对其体外发育率,发育阶段及胚泡大小进行观察。本实验徒手切割小鼠2细胞和8细胞阶段胚胎342枚,切割后成活的半胚为411枚,半胚成活率为60.08%(411/682)。2细胞和8细胞阶段切割的半胚成活率分别为86.3%(209/242)和45.7%(202/442),两者差异极显著(P<0.01),2细胞阶段切割成活率高于8细胞阶段。半胚体外培养24h,2细胞和8细胞切割的半胚发育率分别为78.14%(118/151)和90.14%(150/166)差异极显著(P<0.01),8细胞期半胚发育率明显高于2细胞期。半胚体外培养发育到胚泡期,由半胚发育成的胚泡其直径为80~100μm。与对照组相近。选择发育正常的胚泡,移植到假孕受体子宫,获得由半胚发育成的仔鼠8只。
