类胡萝卜素和维甲酸对不同雌激素状态乳腺癌细胞增殖的影响 被引量:2

摘要 目的 :通过体外细胞研究 ,观察类胡萝卜素和维甲酸对人乳腺癌细胞的作用。方法 :用噻唑蓝 ( MTT)法观察维甲酸和类胡萝卜素对体外培养的雌激素受体阳性 ( ER+ )乳腺癌细胞 MCF- 7和雌激素受体阴性 ( ER- )乳腺癌细胞 MDA- MB- 4 3 5 S的抑制作用。结果 :发现维甲酸和类胡萝卜素对乳腺癌细胞的增殖抑制表现出剂量、时间和结构效应以及细胞的特异性。其中 β-胡萝卜素对 ER+和 ER-乳腺癌细胞均有很强的抑制作用 ,而维甲酸和其他 4种类胡萝卜素 (玉米黄素、虾青素、角黄素、玉米黄素双棕榈酸酯 )对ER+乳腺癌细胞的抑制作用强于 ER-乳腺癌细胞。结论 :β-胡萝卜素对乳腺癌细胞的抑制机制不同于维甲酸和其他 4种类胡萝卜素 。 Objective:To investigate the effects of carotenoids and retinoic acid(RA) on human breast cancer cells in vitro .Methods:The inhibited effect of carotenoids and retinoic acid(RA) on t h e proliferation of breast cancer cells, either estrogen receptor(ER) positive or negative were studied by MTT assay.Results:It was found that the inhibition of carotenoids and RA on brea st cancer cells depended on dosage, time and molecular structure.β carotene co uld strongly inhibit the proliferation of ER+/- breast cancer cells, retinoic ac id and other carotenoids(zeaxanthin, astaxanthin, canthaxanthin, zeaxanthin dipa lmitate) were more potent in ER positive MCF 7 breast cancer cells than ER ne gative MDA MB 435S breast cancer cells. Conclusion:These results suggest that the mechanism of inhibited actio n of β carotene is different from retinoic acid and other carotenoids, the inh ibition was independent of transforming it into RA.
