摘要
目的 探讨 HBV pre S基因免疫和 h IL - 2与 pre S融合基因免疫诱导 BAL B/ c小鼠体液免疫反应的规律 .方法 将 pre S基因及 h IL 2 - pre S融合基因分别克隆入真核表达载体p CMV4,转染 COS- 7细胞 ,明确有目的基因表达后 ,将重组质粒注射 BAL B/ c小鼠臀部肌肉 ,EL ISA方法检测小鼠血清抗体 .结果 p CMV4- pre S接种组 ,3/ 7小鼠产生抗 pre S2抗体 .p CMV4- h IL2 - pre S接种组有 3/ 7小鼠产生抗 h IL- 2抗体 ,而未检测到抗 pre S2抗体 .结论 p CMV 4- pre S可有效诱导小鼠产生抗体反应 .h IL2 - pre S融合基因接种 BAL B/ c小鼠时 ,可诱导产生抗 h IL-
AIM To investigate the humoral immune responses to the immunization of mouse BALB/c with the HBV preS gene and the hIL2 preS2 fusional gene. METHODS HBV preS gene was cloned into the pUC18 hIL2 plasmid, which was connected with the 3terminal of the hIL 2, then the hIL 2 perS fusional gene were inserted into the eukaryotic expression vector pCMV4. The pCMV4 hIL2 preS and pCMV4 preS were built and expressed transiently in COS 7 cells. After purification, the different plasmids were injected into the muscle of BALB/c, respectively. Serum antibodies from the immunized mice were detected by ELISA. RE SULTS Enzyme cutting identification showed that the hIL2 preS and the preS had been inserted into pCMV4. Three out of seven mice in the group of injection of pCMV4 preS plasmids produced the preS2 antibodies. Three out of seven in the group of pCMV4 hIL2 preS achieved the hIL 2 antibodies, but it could not get the preS2 antibodies in this group. CONCLUSION Immunization of the preS gene is able to induce the response of humoral immunity in mice. Injection of the hIL2 preS fusional gene into the muscle of mice could cause production of hIL 2 antibodies which may have negative effect on preS gene immunization.
