银屑病表皮细胞中热休克蛋白的表达意义

Significance of expression of heat shock proteins in epidermal keratinocytes of psoriatic lesions

目的 研究银屑病患者治疗前后皮损表皮细胞中HSPs的表达情况 .方法 应用 L SAB免疫组织化学法和真彩色医学图像分析系统 ,测定 2 5例银屑病患者皮损、非皮损以及不同治疗方法治愈后表皮细胞中和 6例正常人对照组表皮细胞中 HSP2 7和 HSP70的表达及含量 .结果 HSP2 7和HSP70在正常人表皮细胞中呈基础表达 (HSP2 70 .10 0±0 .0 15 A;HSP70 0 .0 94± 0 .0 18A) ,在银屑病患者皮损中表达很弱或无表达 (HSP2 70 . 0 6 8± 0 . 0 2 8A;HSP70 0 . 0 34±0 .0 14A) ,两者有显著性差异 (P<0 .0 5 ) ;在治愈后表皮中其表达量逐渐恢复 (HSP2 70 .10 5± 0 .0 2 8A;HSP70 0 .0 78±0 .0 2 2 A) ;而不同治疗方法治愈后其表皮细胞表达含量间无明显差异 .结论 在银屑病患者皮损中 ,HSP2 7和 HSP70表达消失 ,随着皮损治愈 ,HSP2 7和 HSP70表达逐渐恢复 ,提示HSP2 7和 HSP70可能在银屑病应激保护机制中发挥着重要作用 .

AIM To study the expressions of HSPs in epidermal keratinocytes of pre and post treatmental psoriatic lesions. METHODS We detected the expressions of HSP27 and HSP70 in epidermal keratinocytes of pre and post treatmental psoriatic lesions (25 cases) and normal control skins (6 cases) by means of immunohistochemical LSAB method and the Color Medical Image Analysis System (CMIAS). RESULTS HSP27 and HSP70 were expressed constitutively in epidermal keratinocytes of nomal skins (HSP27 0.100± 0.015 A ; HSP70 0.094±0.018 A ). In epidermal keratinocytes of psoriatic lesions, the expressions of HSP27 and HSP70 were very weak, and even negative (HSP27 0.068±0.028 A ; HSP70 0.034±0.014 A ). The differences were significant between 2 groups ( P <0.05). The expressions of HSPs were increased gradually in epidermal keratinocytes after treatment (HSP27 0.105±0.028 A ; HSP70 0.078± 0.022 A ). But the differences of expression of HSPs in epidermal keratinocytes were not significant among different therapies. CONCLUSION Expressions of HSP27 and HSP70 are negative in epidermal keratinocytes of psoriatic lesions, increased gradually with treatment. HSPs may play an important role in stress protective mechanism of psoriasis.
