摘要 目的 :探讨炎性细胞因子对肝脏胆汁酸分泌机制的影响。 方法 :利用体外原代培养大鼠肝细胞、枯否细胞(Kupffercell,KC)的方法和反向高效液相色谱 (RP -HPLC)技术 ,观察部分炎性细胞因子 (IL 1、IL 6、TNFα)以及被内毒素 (LPS)活化的KC上清液对肝细胞胆汁酸分泌的影响。 结果 :(1)炎性细胞因子和活化的KC上清液刺激后 ,肝细胞培养上清液中GCDCA、GCA和TGA三种结合胆汁酸含量均较对照组明显降低 (P <0 0 1) ;(2 )实验组 (GCA±GCDCA) /TGA(G/T)的比值较对照组G/T的比值明显升高 (P <0 0 5 )。 结论 :炎性细胞因子可以改变大鼠肝细胞胆汁酸的分泌机制 ,导致胆汁酸分泌含量的减低 。 To investigate the effects of cytokines on bile acid secretion of hepatocytesMethods:The technique of primary culture of hepatocytes and Kupffer cells in vitro and the techique of RP-HPLC were used.Some components of bile acid in the supernate of cultured hepatocytes were measured after the hepatocytes were stimulated with an array kinds of cytokines(IL-1,IL-6,TNF-α)and the supernate of Kupffer cells activated by LPS. Results:The concentrations of GCDCA,GCA and TGA in the supernate of cultured hepatocytes stimulated by cytokines and supernate of culture Kuppffer cells activated with LPS were significantly lower in experimental group than in control group(P<0.01).The ratio of (GCA±GCDCA)/TGA of experimental group is significantly higher than that of control group (P<0.05).Conclusion:The bile acid secretion can be changed by cytokines and activated Kupffer cells,thereby bile acid secretion be reduced and the bile acid components be changed.
