摘要
以 7年生落叶松杂种 (长白落叶松×日本落叶松 ,Larixolgensis×L .kaempferi)无性系幼树的当年生半木质化嫩枝为材料进行扦插。利用间接酶联免疫吸附测定法 (ELISA)分析了生根率高、中、低 3组无性系插穗在生根过程中 4种内源激素 (IAA、ABA、ZR、GA4 )的动态变化 ,探讨了插穗生根与内源激素的关系。结果表明 :生根率高的 3号无性系插穗 ,在扦插初期ABA含量低 ,而生根率中等和生根率很低的两组无性系ABA含量高 ,ABA对扦插生根有抑制作用 ;在生根率较高的 3号无性系插穗生根过程中 ,ZR的含量始终比生根率中等和生根率低的两组无性系的含量高 ,对插穗生根起主要作用 ;GA4 在生根率高的 3号无性系根原基形成期的含量比其它两组的含量都高 ,GA4 对根原基分化起重要作用 ;IAA在 3组无性系中的含量变化不明显 ,看不出对生根起促进作用 。
The softwood cuttings from larch hybrid( Larix olgensis × L . kaempferi ) in 7 years old have been propagated. During the cuttings rooting, four endogenous hormones, namely, auxin(IAA), abscisic acid (ABA), cytokinin (ZR) and gibberelin4 (GA 4) within the cuttings from three clones in different rooting rate were analyzed by means of the enzyme-linked immunosorbent assays (ELISA). The purpose was to analyze the relationship between the endogenous hormones and rooting cutting. The results showed that the cuttings from No.3 clone get the highest rooting ability among three clones; the ABA concentration in clone No.3 within the highest rooting rate is lower than the other two clones, especially during the early stage of cuttings, and visa versa for the other two clones. That means ABA concentration within the plants having some constraint effect on the cuttings rooting. The concentrations of ZR and GA 4 in No.3 clone are higher than those of the other two clones. It can be seen that ZR concentration in the cuttings keeps higher level during its all rooting time, and GA 4 concentration in the cuttings is just higher during the differentiation of root primordium. There is no clear change for IAA concentration in the cuttings from all clones during cuttings rooting.
