Cytotoxicity and Cell Cycle Disturbance Effects of Pteris Semipinnata L.Extraction on HL-60 Cells

目的 :研究蕨类植物半边旗提取物对HL 60细胞的体外杀伤作用及对细胞周期的影响。方法 :用噻唑蓝 (MTT)法测定细胞成活率 ;用流式细胞术测定细胞周期时相分布。结果 :半边旗乙醇总提取物PSE及纯化合物 5F、6F及A对HL 60细胞有强烈的杀伤作用 ,具明显的时间和剂量效应 ;PSE、5F、6F及A作用HL 60细胞 2 4h后 ,G0 /G1期细胞比例明显下降 ,S期细胞比例明显升高 ,G2 /M期轻度升高 ( 5F作用组例外 )。结论 :半边旗提取物对HL 60细胞有强烈的杀伤作用 ,明显影响细胞周期时相分布。

Objective To investigate the cytotoxicity and cell cycle disturbance effects of extraction from Pteris Semipinnata L.(PSL)on HL 60 cells in vitro.Methods Cytotoxicity assay was tested by tetrazolium salt(MTT) method.Cell cycle was determined using Flow Cytometry (FCM) analysis.Results The total ethanolic extraction (named as PSE)of PSL,compound 5F,6F and A showed strong cytotoxicity against HL 60 cells in a obvious time and dose dependent manner.After exposure of HL 60 cells to PSE,compound 5F,6F and A for 24 hours,the cell cycle distribution was changed compared to control:the percentage of cells in G 0/G 1 phase decreased while increased in S phase significantly,and G 2/M phase ratio increased slightly except the group of compound 5F treatment.Conclusion Extraction from PSL showed strong cytotoxicity and cell cycle disterbance effects on HL 60 cells.
