摘要
目的 通过观测东莨菪碱对雄性SD大鼠吗啡位置偏爱 (相当于渴求 )形成过程的影响 ,以期揭示胆碱能M型受体参与吗啡精神依赖的形成。方法 将 2 4只无自然地点偏爱的雄性SD大鼠采用同窝异体配对设计随机分为实验组与对照组 ,均在建立吗啡依赖模型的过程中 (10天 )及戒断后 (共 14天 )进行 8次吗啡位置偏爱实验 ,比较两组大鼠在吗啡侧的停留时间。其中实验组于注射吗啡时加注东莨菪碱。结果 在吗啡依赖形成及戒断过程中 ,实验组 (吗啡 +东莨菪碱组 )和对照组 (吗啡 +生理盐水组 )在吗啡侧的停留时间均长于生理盐水侧 ,产生了对吗啡的位置偏爱。但两组相比实验组大鼠在吗啡侧的停留时间显著短于对照组 (P <0 .0 5 )。结论 东莨菪碱明显抑制了大鼠吗啡位置偏好的形成 。
Objective To explore the role of cholinergic(M ) receptors in the development of morphine psychological dependence from observa tion of the action of scopolamine on morphine-induced conditioned place prefere nce (CPP, craving). Methods 24 male Sprague-Dawlay rats were randomly assigned to experimental or paired control group. Morphine depende nt animal model was developed by i.p. morphine twice a day for 10 days in ascend dosage schedule and the CPP was also deve loped at the same time. scopolamine (hyoscine) was administered intraperitoneall y accompanying with morphine to experimental group. Morphine treatment discontin ued on day 11. CPP was recorded in the two groups in the period of dependent dev elopment and withdrawal to assess impact of scopolamine. Results There was signi ficant difference (P<0.05) between the experimental and control group in CP P in the period of dependent development and withdrawal. Conclusion scopolamine att enuated the development of CPP significantly cholinergic (M) receptors play a po ssible role in morphine psychological dependence in SD rat.
