摘要
目的 :检测任意皮瓣移植时 ,皮瓣局部组织中的血小板激活因子 (PAF)水平 ,探讨PAF及其受体拮抗剂(BN5 2 0 2 1)在移植皮瓣成活过程中的作用。方法 :实验 1:皮瓣移植术后 0、2、4、6、8h时将皮瓣取下 ,分蒂段、中段、末段匀浆提取组织液 ,检测血小板激活因子前体 (lyso PAF)的水平变化。实验 2 :皮瓣移植术前 30min ,皮瓣局部皮下注射单一剂量的银杏苦内酯B(BN5 2 0 2 1) ,术后 7d观察皮瓣成活情况。结果 :①皮瓣组织中的lyso PAF含量明显增高 ,尤以皮瓣中段为明显 ;②单一剂量的BN5 2 0 2 1明显地提高皮瓣的成活面积 (5 9.8%比 31.8% ,P <0 .0 0 1)。结论 :①鼠任意皮瓣移植时 ,皮瓣组织中的PAF含量明显增高 ;②单一剂量的BN5 2 0 2 1可以明显地增强任意皮瓣的成活能力 ;
Objective: To examine the level of platelet activating factor in the skin flap tissue, to evaluate the possible benefit and to investigate the effect of platelet activating factor antagonism in the survival of random flaps. Methods: Experiment 1: 0, 2, 4, 6, 8 hours after the transplantation of the bilateral flaps, biopsies of the 60 SD rats were collected sequentially and the lyso PAF level in the flaps were examined. Experiment 2: A lipophilic platelet activating factor receptor antagonist (BN52021) was administered in a single dose to treat flaps by a local percutaneous injection route 30 min prior to the operation. 7 days later, the survival of skin flaps were investigated. Results: ①The lyso PAF level in the flaps were found to be higher than that in normal tissues; ②The platelet activating factor receptor antagonism augmented the survival of skin flaps significantly (59.8% versus 31.8%, P<0.001). Conclusion: ①The platelet activating factor level in random flaps subjected to operations and ischemia was found to be higher than that in normal tissues; ②The specific platelet activating factor receptor antagonist BN52021 is beneficial to the survival of skin flaps; ③ During transplantation, the platelet activating factor level is related to the survival of random flaps.
