ESTABLISHMENT OF NEW ZEALAND RABBIT OSTEOPOROTIC MODEL BY OVARIECTOMY

摘要 目的 摘除新西兰兔双侧卵巢 (去势法 ) ,观察其骨骼、碱性磷酸酶、体重等的改变情况 ,探讨去势法建立新西兰兔骨质疏松症模型的有关问题。方法 摘除 5月龄新西兰兔双侧卵巢 ,3个月后测定其胫骨骨密度和骨皮质厚度、股骨生物力学性能、碱性磷酸酶、胫骨组织学特征等。结果 去势法 3个月后 ,各兔胫骨骨密度明显比正常对照组的低 ( P<0 .0 1) ,骨皮质厚度也明显比正常对照组薄 ( P<0 .0 5 ) ,股骨生物力学性能出现了明显的退行性改变 ,胫骨组织学显示骨小梁稀疏、排列紊乱 ,碱性磷酸略有升高 ,体重较正常组明显增加。结论 去势法建立新西兰兔骨质疏松症模型切实可行 ,造模时间 3个月即可。 Objective To explore whether rabbit osteoporotic model could be established by ovariectomy, New Zealand rabbits' ovaries were ectomized and the characteristics of their tibias and femora were studied.Methods Ten New Zealand rabbits were divided into two groups equally, which were operation group (OG) and shame operation group (SOG). The bilateral ovaries of the OG were ectomized, while those of the SOG were not ectomized. Three months later, the histological and machinanic characteristics of the tibias and femora were studied, the weights of the bodies and uteri were measured, the tibial bone mineral density (BMD) and thickness, alkaline phosphatase were measured.Result Although all the weights of the bodies had increased, these of the OG had increased more greatly than the SOG(P<0.05). There were significant differences between the two group's tibial BMD and thickness (P<0.01, P<0.05). The biomechanical characteristics of the femora in the OG had apparent osteoporotic changes. The histological characteristics showed that the cortical bones of the OG were thinner and more porous than those of the SOG.Conclusions These results suggest that ovariectomy is a good method to establish New Zealand rabbit model of osteoporosis, the rabbits should be five months old and three months are enough to establish the model.
