摘要
研究当归对大鼠肝脏低温保存效果的影响及可能的作用机制。方法:采用大鼠肝脏低温保存模型,利用生化分析、光镜及电镜技术测定肝脏功能及观察肝脏组织形态学改变。结果:各保存时限(12h、24h、36h),B组(25%当归注射液20ml/CMU-1保存液1L,大鼠肝脏置于其中保存)和C组(25%当归注射液40ml/CMU-1保存液1L,大鼠肝脏置于其中保存)大鼠肝脏灌洗液的AST,LDH水于均较A组(单纯CMU-1保存液组,大鼠肝脏置于其中保存)为低,差异有统计学意义(P<0.95),B组和C组间差异不显著(P>0.05)。在各保存时限内,B组和C组肝细胞和肝窦内皮细胞的损伤均较A组为轻,B组和C组损伤程度相当。结论:当归对低温保存的大鼠肝脏有保护作用;当归在保存液中达到一定浓度后(B组),即使再增加浓度(C组),并不能明显增加其保护作用。
To study the influence of Angelicae Sinensis upon the rat liver during cold preservation. Methods: The rat liver model of cold preservation was set up, measured activities of AST and LDH in the perfusate, and observed pathohistorical changes in different groups from microscopy and electronmicroscopy. Result: In different preservation times, the activities of AST and LDH of B group and C group was noticeably lower than A group' s (P<0. 05), the damage of hepatocytes and endothelial cells of B group and C group was slighter than A group' s, the damage of B group and C group was similar. A group (control group) was simple CMU-1 solution preservation group B group (experimental group one ) was addition of normal dose of Angelicae Sinensis into CMU-1 solution, the ratio of 25 % Angelicas Sinensis to CMU-1 solution was 20ml/lL: C group (experimental group two) , the ratio of 25% Angelicae Sinensis to CMU-solu-tion was 40ml/lL. Conclusion: The results indicate that Angelicae Sinensis has protective effects upon the rat liver during cold preservation. When Angelicae Sinensis arrived effective concentration in B group, though enhancing concentration in C group, the protective effects don't accrue.
