粉防己碱对鼠脑缺血海马神经细胞的保护作用 被引量:5

Effects of Tetrandrine on Intracelluar Free Ca^(2+ ) and Lipid Peroxidation of Hippocampus in Rat Cerebral Ischemic Damage

摘要 采用全脑缺血再灌注模型 ,应用 Fura- 2荧光探针检测海马神经细胞游离钙 ([Ca2 + ]i)水平 ,硫代巴比妥显色法检测海马匀浆中过氧化脂质 (L PO)含量 ,同时电镜下观察海马 CA1 区神经细胞改变 ,以评价药物粉防己碱的作用。结果表明 :粉防己碱预处理后 ,能够显著抑制全脑缺血 2 0 min再灌注 6 0 min海马组织 [Ca2 + ]i及 L PO含量的升高 ,减轻神经细胞病理损伤。 The cerebral model of ischemia for 20 min and reperfusion for 60 min was used. Fluorescent probe Fura 2 of Ca 2+ indicator and thiobarbituric acidreactive (TBA) method were applied to measure the levels of intracelluar free Ca 2+ and the content of lipid peroxidation (LPO) respectively. The neuronal cell changes of CA 1 area at hippocampus were observed under an electron microscope to evaluate the effects of tetrandrine. The results showed that the levels of intracelluar free Ca 2+ and LPO contents in the tetrandrine pretreated group were significantly lower than in the saline pretreated group respectively, and the pathological damage to the neuronal cells was alleviated in the tetrandrine pretreated group. It was suggested that tetrandrine had a protection on brain to some extent.
