摘要
为了从细胞水平探讨近交系小鼠对胰岛素 ( Ins)的敏感性 ,测试了 3种近交系小鼠脂肪细胞在 Ins刺激下的葡萄糖转运和 Ins抑制异丙肾上腺素介导的脂肪细胞分解。结果显示 :A/ J小鼠脂肪细胞对葡萄糖的转运显著高于Balb/ c J和 12 9/ J小鼠 (分别为 P<0 .0 5和 P<0 .0 1) ,而 Balb/ c J又显著高于 12 9/ J( P<0 .0 5 ) ;Ins对 A/ J小鼠脂肪细胞脂解的抑制作用显著高于 Balb/ c J和 12 9/ J( P<0 .0 1)。表明 A/ J小鼠脂肪细胞对 Ins敏感性高 ,Balb/ c J次之 ,12 9/J较低。这对研究不同种系的近交系小鼠的生理病理特征及是否易于发生与胰岛素抵抗有关的疾病将提供一定的依据。
In order to study the insulin sensitivity of inbred mice at cellular level, glucose transport and lipolysis response to insulin were determined in adipocytes isolated from three strains of mice. The results showed that the insulin-stimulated glucose transport of adipocytes from A/J mice was obviously higher than that from both Balb/cJ and 129/J mice (P<0 05 and P<0 01, respectively), whereas glucose transport of adipocytes from Balb/cJ mice was higher than that of 129/J mice (P<0 05). The ability of insulin to inhibit isoproterenol-induced lipolysis in A/J mice was obviously higher than in both Balb/cJ and 129/J mice (P<0 01). The results demonstrated that the adipocytes of inbred mice showed various sensitivities to the action of insulin: A/J mice with the highest sensitivity, Balb/cJ mice moderate and 129/J mice lowest, which would provide physiological and pathological trait of inbred strains and is worth studying which strain of mouse is susceptible to develop disease related to insulin resistance.
