摘要
目的 :本实验采用几种醇改性戊二醛处理后的牛心包 ,希望寻找一种理想的生物瓣处理方法。方法 :分为五组 ,即新鲜对照组及戊二醛、2 ,3 -丁三醇、1,3 -丙二醇、1,2 -丙二醇组。分组将牛心包试片植入SD大鼠背部皮下 6 0d后 ,行组织钙定量测定及组织学检测。结果 :2 ,3 -丁二醇改性后的牛心包试片钙平均含量为3.99± 0 .96ug/mg干重 ,与GA组、1,2 -丙二醇 ,1,3-丙二醇试片的钙平均含量 (2 41.89± 7.79、2 44 .2 6± 6 .39、16 7.32± 12 .2 9ug/mg干重 )相比 ,钙化 ,钙化明显减轻 (均P <0 .0 1) ,但与对照组 (4.11± 2 .2 3ug/mg干重 )相比无显著性差异 (P >0 .0 5 )。病理组织学检查显示 2 ,3 -丁二醇组和 1,3 -丙二醇组试片仅有轻度形态学改变。而GA组和 1,2 -丙二醇组试片有严重的胶原变性 ,大片钙化。结论 :戊二醛鞣制的生物材料经2 ,3 -丁二醇改性后 。
Objective:To find a new ideal fixative for the post-treatment of the bovine pericardium tanned with glutaraldehyde-(G-tBP) to reduce calcification and to improve durability of the bioprostheses.Methods:Fresh bovine pericardium was tanned with 0.625% glutaraldehyde (GA) for 1 week,then post-treated in one of four ways for 60 days:(1)Group A:2,3-butanediol;(2)Group B:1,3-propandiol;(3)Group C:1,2-propandiol;(4) Group D:0.3% GA and Group E:Fresh bovine pericardium.Treated bovine pericardium was cut into 1 cm 2 pieces and washed for 30 minutes with normal saline before implantaiton.One piece of each group was implanted subcutaneously in one of weanling SD rats,total 10 animals.After sixty days,all the rats were sacrificed,the retrieved specimens were examined grossly,and calcium analysis was determinated by flame atomic absorption spectrophotometer,histologic examination and election microscopy were also carried out.Results:After 60 days implantation,the results showed that the 2,3-butanediol post-treated bovine pericardium(group C)and fresh bovine pericardium (group E)had less calcification (calcium 3.99±0.96 and 4.11 ±2.23ug/mg dry weight respectively)than other groups:gourp A(calcium 244.26±6.39ug/mg dry weight),group B(calcium 167.32±12.29ug/mg dry weight)and group D (calcium 241.89±7.75ug/mg dry weight)(P<0.01).Pathohistologic examinations demonstrated a prominent inflammatory response in the form of giant cells and mononuclear cells infiltration associated with intrinsic collagen disruption in the G-tBP;the 2,3-butanediol post-treated bovine pericardium maintained morphologic integrity with only mild inflammatory response. Conclusions:G-tBP post-treated with the 2,3-butanediol appears less calcification.
