子宫颈癌端粒酶活性与DNA含量的关系 被引量:2

摘要 目的 :探讨端粒酶活性与宫颈癌的相关性以及与DNA含量的关系。方法 :采用PCR TRAP方法对 2 8例宫颈癌组织 ,10例正常宫颈组织 ,6例宫颈不典型增生组织的端粒酶活性进行检测。采用流式细胞仪检测肿瘤细胞DNA含量。结果 :宫颈癌、宫颈不典型增生组织端粒酶活性表达明显高于正常宫颈组织。宫颈癌端粒酶活性表达组DNA异倍体率、SPF、DI、PI平均值明显高于端粒酶阴性组。结论 :端粒酶的激活在宫颈癌起源过程中可能是较早的事件并且和其发生发展有关。 Objective To investigate the possible relationship between telomerase activity and progression of cervical carcinoma.To define the correlation between telomerase activity and cellular DNA content in cervical carcinoma.Methods Telomerase activity in 28 specimens of cervical carcinoma,6 specimens of cervical dysplasia and 10 specimens of normal cervical tissues were detected by PCR based sliver staining Telomeric repeat Amplification Protocal (TRAP).Flow cytometry was used to detect cellular DNA content,ploidy state and portions of cells in each phase of the cell cycle.Results Telomerase activity in cervical carcinoma and in dysplasia is significantly higher than that in normal cervical tissue.DNA aneuploidy cervical carcinoma was correlated with unfavorable characteristics and telomerase activity.Conclusion Telomerase activity might contribute to the progression of cervical carcinoma and might be a relatively early event.
