苦参促粒系造血机制及对白血病HL-60细胞的诱导凋亡作用 被引量:2

摘要 目的 :进一步探讨苦参促粒系造血机制及新的抗白血病作用机理。方法 :建立实验性环磷酰胺 (Cy :0 .0 8g/kg)低白细胞血症小鼠模型 ,观察苦参对粒系造血的影响和对白血病HL - 6 0细胞的诱导凋亡作用。结果 :苦参对Cy所致外周血白细胞和中性粒细胞数下降具有对抗和促恢复作用 (P <0 .0 1) ,能增加骨髓有核细胞、粒 -巨噬系祖细胞 (CFU -GM)数 (P <0 .0 1)促进CFU -GM增殖并诱导其向粒系、巨噬系分化 ,且其促粒系造血作用与鲨肝醇一致。此外 ,苦参可明显诱导白血病HL - 6 0细胞凋亡 ,呈药物浓度 -效应关系。结论 Objective:To further study the effects of Sophora flavescens(SF) on granulopoiesis and apoptosis-induced in HL-60 cells.Methods:Experimental leukopenic mice model induced by cyclophosphamide (Cy:0.07g/kg)and apoptosis characteristic DNA fragment assay were established,respecively.Results:The results showed that SF (2.9kg -1 .d -1 ×6d) had antagonized the decrement of the number of white blood cells (WBC),neutril cells (NC) in peripheral blood and that of bone marrow nucleated cells (BMNC),CFU-GM in bone marrow,stimulated proliferation and induced differentiation towards macrophages of CFU-GM.In particular,leukogenic effect of SF was approximately batiolum.Furthermore,5~20μg/ul of SF significantly induced apoptosis in HL-60 cells in a dose-dependent manner.Conclusions:SF may be used to prevent and treat granulopoiesis suppression caused by chemotherapy,and exert its anticancer effect by means of inducing tumor cell apoptosis.
