凤丹的体外增殖研究

摘要 为探索凤丹体外增殖的最佳条件 ,利用组织培养技术进行观察。结果显示以下培养条件较适宜 :凤丹种子灭菌后在 35℃下于摇床上振动 72h ,取出胚 ,在 1/ 2MS +10 %椰子汁的培养基上培养 1个月 ,然后将带有腋芽的茎段切下置于MS +BA(1mg/L)培养基上进行培养 ,形成丛生芽 ,当芽长出 2~ 3片叶子时 ,转移至MS +IBA(1mg/L) +3 %蔗糖培养基上 ,1个月后即长出白色的直根 。 In order to explore the best condition for external culture of Fengdan,tissue culture technology was used for observation.The results showed that the following culture condition was appropriate.After disinfection,the fengdan seeds are vibrated for 72 hours on the shaking bed at the temperature of 35℃.Embryo is taken out and cultured on the medium of 1/2MS+10% coconut juice for one month.Then,the stem with bud is cut down and cultured on the medium of MS+BA(1mg/L)to form thickly growing sprouts.When 2~3 leaves grow out,the sprouts are moved to be cultured on the medium of MS+IBA(1mg/L)+3% sucrose.One month later,white straight root grows out to form a complete plant.
