摘要
目的 :研究碱性成纤维细胞生长因子 (basicfibroblastgrowthfacfor ,bFGF)对体外培养脊髓神经元的作用。方法 :取E14 大鼠脊髓神经组织 ,进行体外原代培养 ,分别加入不同浓度的bFGF。在相差显微镜下观察细胞生长发育情况 ,并测定其细胞内琥珀酸脱氢酶的活性。实验结果用t检验分析。结果 :bFGF处理组神经元密度明显高于对照组 ,神经元胞体大而饱满 ,突起较长。细胞内琥珀酸脱氢酶活性较高 ,培养 2 0d后神经元存活的数量也明显高于对照组。结论 :bFGF能促进脊髓神经元在体外培养早期的存活 ,增加神经元的胞体直径和突起长度 ,增强神经元细胞内琥珀酸脱氢酶的活性 。
Objective:To study the effects of bFGF on cultured spinal cord neurons.Method:Spinal cord neurons from embryonic day 14 rat were primarily cultured with different concentrations of bFGF,The growth condition of cell was observed with phase contract microscope,and intracelluer dehydrase activity was detected.Result:The neuron number and body diameter of bFGF treated groups were significantly larger with longer processes and elevated dehydrase activities. bFGF enhenced the neuron survival rate of 20th day to a significant degree.Conclusion:bFGF could enhence the survival of cultured embryonic spinal cord neurons,and increase their body diameter and process outgrowth,and dehydrase activity.
