摘要
目的 建立简便、灵敏 ,试剂稳定的 Xylidyl Blue 血清镁测定方法。方法 用 Xylidyl Blue 作显色剂 ,EGTA作钙掩蔽剂 ,选用甘氨酸 - Na OH作缓冲体系。用分光光度法测定血清镁。结果 线性范围0~ 3.35 mmol/L,精密度 :批内 CV<1 .67% ,总 CV<2 .2 8% ,平均回收率为 99.8% ,与 Camagite( X)比较 :r=0 .983,Y=1 .0 0 2 X- 0 .0 4 5 ,与甲基百里酚蓝 ( MTB) ( z)比较 :r=0 .995 ,Y=0 .965 Z+ 0 .0 4 8,测试的灵敏度分别是上述方法的 2倍和 4倍。结论 该法简便、灵敏 ,试剂稳定性明显优于 Calmagite法和MTB法 。
Aim To establish a simple sensitive and stable method for determination of magnesium in serum by singleness reagent with Xylidyl blue I.Method Xylisyl blue I was used as a coloring reagent and EGTA as a reagent covering calcium in the glycine-NaOH buffer system to determine magesium by spectrophotometry.Results The linearity was 0~3.35 mmol/L;The precision were<1 67% for within-run CV,and<2.28% for total CV.The average rate of recovery was 99.8%.Correlation between results obtained by this method(Y) and Calamagite(X) was Y=1 002X-0.045,r=0.983 and MTB(Z) was Y=0.965Z+0.048,r=0.995 The method was 2 time the sensibility of Calmagite and 4 time the sensibility of MTB.Conclusion The method is simple sensitive and stable for routine or automatic measurement of magne-sium in serum.
