建立结核分枝杆菌耐药基因的检测方法

摘要 目的 了解耐药结核分枝杆菌基因突变情况 ,建立结核分枝杆菌耐药基因的检测方法。方法 1 0 8例临床痰标本分离株均做 PCR- SSCP和传统梯度药敏试验。结果 耐 SM( rpsl)、RFP( rpo B)、INH( kat G)基因突变率分别为 78.5 % ,78.2 % ,70 .5 %。其中高耐 SM、REP、INH分离株突变率分别为86.9% ,89.3% ,84.3%。低耐分离株突变率分别为 2 8.5 % ,1 6.6% ,7.1 %。结论 结核分枝杆菌耐药基因突变与耐药水平联系是密切的 ,且结核耐药分枝杆菌基因突变绝大多数易发生在高耐药株中 ,也有少部分在低耐药株发生突变。 Aim To understand the relationship between drug-resistant gene mutation and drug-resistant level in my-cobacterium tuberculosis.Methods 108 clinical isolates from spurum specimens were analyzed by PCR-SSCP technique and traditional drug susceptibility tests.Results The gene mutation rate of SM,RFP and INH resistawt isolates was respectively 78.5%,78.2% and 70.5%.The mutation rate of SM,RFP and INH higher contentration resistant isolates was respectively 86.9%,89.3% and 84.3%.But 28.5%,16.6% and 7.1% in lower concentration resistant isolates.Conclusion The relationship is intimate between the gene mutation and drug resistant level in resistant Mycobacterium tuberculosis. The gene mutation mostly occured in higher concentration resistant isolates,only the minority is occured in lower concentration resistant isolates.
