对Goldblatt大鼠心脏收缩功能的研究及Valsatan对心肌收缩功能影响 被引量:5

摘要 目的 本文应用超声研究Goldblatt大鼠心脏收缩功能的改变 ,并应用Valsartan(AT1受体拮抗剂 )治疗 ,观察其对心功能的影响。方法  34只SD大鼠 6~ 8周龄 ,分为Valsartan治疗组 (n =l3,V组 ) ,非Valsartan治疗组 (n =11,NV组 )和假手术组 (n =10 ,S组 ) ,V组和NV组在左肾动脉置入内径为 0 .3mm的银夹 (2Klc) ,每周测一次血压 ,每 2周测一次超声心动图 ,并计算左室收缩功能 ,术后 2周 ,V组给予Valsartan 30mg/kg/d一次性灌胃治疗 ,S组和NV组每天用饮用水灌胃 ,共 10周。结果  (1)各组大鼠治疗前一般情况和左室功能 (EF ,FS、mFS)无差异 ;(2 )与NV组比较 ,术后 10周V组尾动脉压降低 75 .6mmHg ;术后 10周NV组相对室壁厚度、左室重量、左室重量指数明显增加 ,与V组和S组比较P均小与 0 .0 5 ,有显著意义 :NV组舒张期左室内径明显变小 ,NV组 <S组 <V组与S组和V组比较有意义 ,而S组和V组比较无意义 ;(3)NV组的左室收缩功能明显下降 ,其中射血分数 (EF)、左室中层缩短率(mFS)、实测 /预测mFS与V组实测 /预测mFS和S组实测 /预测mFS比较均有显著差异 ,而S组与V组比较无差异 ;但各组问的FS和预测mFS均无显著差异 ;(4 )相关分析 ,各组大鼠实测 /预测mFS与左室腔舒张内径成正相关 (r=0 .6 12 ,P <0 .0 0 0 1) ; Objective To study left ventricular performance in Goldblatt rat and the effect of angiotension Ⅱ receptor antagioists Valsartan on left ventricular function. Methods 34 Males Spragne Dawley rats (6~8 weeks old) were studied. The rats were divided into three groups: valsartan treatment group (V group, n =13), non valsartan treatment group (NV group, n =11) and sham operation group(S group, n =10). In the group V and NV, a silver clip(two kidney,one clip ) with 0 3 mm lumen was placed on the left renal artery. A sham operation was performed in the rats of show group. The systolic blood pressure and M mode echocardiography were examined weekly. The left ventricular functions were calculated. Valsartan was dissolved in drinking water and given to group V at the dose of 30 mg/kg body weight per day. Results (1)The blood pressure,relative wall thickness,left ventricular weight,left ventricular index in group NV were increased significantly after 10 weeks of hypertension compared with group V and group S ( P <0 05). (2)Left ventricular functions in group NV (EF,mFS,observed/preadicted mFS)decreased significantly compareing with group V and group S ( P < 0 05 ),But there were no significant changes in FS and perdicted mFS in any groups. (3) Observed/predicted mFS was positive correlated with left ventricular diastolic dimension ( r =0 612, P < 0 0001 ); Relatived wall thickness was negatively related to MESS ( r =0 563, P < 0 001 ). Conclusion (1) Hypertension and systolic dysfunction can be developed in 2K1C rats. (2) mFS is better than FS in estimating the left ventricular performance. (3) Increasing of wall thinckness especially concentric hypertrophy resulted in deterioration of ventricular function. (4) Valsartan can reduce the regression of LVH and improve the left ventricular function in 2K1C rats.
