摘要
目的 :探讨端粒酶活性在乳腺癌诊断中的临床意义并分析端粒酶表达与P53 基因突变的关系。方法 :采用TRAP -Hyb以及PCR/SSCP法检测乳腺肿块中端粒酶的活性和P53 基因突变。结果 :乳腺癌标本端粒酶的阳性率 88.1% ,乳腺良性病变标本端粒酶的阳性率 5 .7% ,两者差异显著 (P <0 .0 0 5 )。端粒酶表达和肿瘤大小、淋巴结转移无相关性。P53 基因突变率在乳腺癌标本中为 85 .7% ,在乳腺良性病变标本中为 2 .9% ,两者差异显著 (P <0 .0 0 5 )。结论 :端粒酶活性在乳腺组织中可作为一种新的肿瘤标记物 ,在乳腺癌诊断中具有一定应用价值。其与P53 基因突变有关 ,推测P53
Object:To study the clinical significance of telomerase activity in the diagnosis of breast cancer and association with P 53 gene mutation. Methods: TRAP-Hyb and PCR/SSCP were used to detect the telomerase activity and P 53 gene mutation in breast masses. Result: Telomerase was positive in breast cancer specimens with positive rate of 88.1%, while it was positive in benign breast diseases with positive rate of 5.7%. Positive rates were significant different between the two groups ( P <0.005). Telomerase activity did not correlate with tumor size and lymph node metastasis. However, it correlated with P 53 gene mutation ( P <0.005). P 53 gene mutation rate in breast cancer tissues (85.7%) was significantly higher than that in benign breast tissues (2.9%). Conclusion: As one new tumor marker, telomerase activity is useful in the diagnosis of breast cancer, which is correlated with P 53 gene mutation. P 53 gene mutation may be one of the reasons of telomerase activated.
