摘要
目的 从肺癌患者外周血诱导扩增的树突状细胞 ,观察其形态、表型、抗原递呈功能和对LAK细胞抗肿瘤作用的影响。方法 肺癌患者外周血单个核细胞经GM -CSF、IL - 4诱导扩增成为树突状细胞 ,FACS检测其表型 ,MLR测定其抗原递呈能力及对LAK细胞增殖的影响。TAA冲击的DC与LAK细胞混合培养诱导的CTL ,用MTT法测定其细胞毒。结果 肺癌患乾外周血诱导的树突状细胞具有典型的形态、表型特征和激发MLR的能力 ,TAA冲击的DC能显著地提高LAK细胞的增殖能力和增强对靶抗原细胞的细胞毒效应。结论 从肺癌患者外周血中可获得典型的树突状细胞 ,经TAA冲击的DC能从LAK细胞中诱导出特异性CTL细胞 ,以该DC为基础的疫苗联合LAK细胞 。
Objective To induce and proliferate dendritic cells from the peripheral blood of lung cancer patient and to observe the morphologic and phenotypic function of antigen presenting as well as the effect of antitumor activity on the LAK cells. Methods Granulocyte/macrophage colony stimulating factor and inteleukin 4 were used to cultivate DC from peripheral blood mononucleu cells (PBMC) of lung cancer patients. The phenotypes of those cells were observed with FACS. Their abilities of antigen presenting and effect of proliferation of LAK cells were determined by MLR. The cytotoxic T lymphocyte (CTL) were induced by using a co-culture method with DC pulsed by TAA and the LAK cells, and then cytotoxin effect of CTL was tested by MTT. Results The DC induced from PBMC of lung cancer patient had typical morphologic and phonotypic characteristics and the ability of potent stimulators in the mixed leukocyte reaction. The DC impulsed with TAA can markedly increase proliferation of the LAK cells and enhance specific cytotoxin effect. Conclusion These typical dendritic cells cultured from the PBMC of lung cancer patient were impulsed by TAA, which is able to induce specific CTL from the LAK cells. Integrating the dendritic cells vaccine with the LAK cells may play an important role in lung cancer biotherapy in vivo.
