摘要
目的 探讨异丙酚对大鼠血流动力学和血浆内皮素 (ET)、降钙素基因相关肽 (CGRP)的影响。方法 12~ 14周 Wistar大鼠 2 4只随机分为三组 ,每组 8只。A组 (对照组 ,注射 0 .9%生理盐水 )、B组 (低剂量异丙酚 )和C组 (高剂量异丙酚 )静注异丙酚 5 mg/ kg诱导麻醉后分别静脉持续注入异丙酚 30、 6 0 m g· kg- 1 · h- 1 3 0 min。总容量均相同 ,记录不同时间性的收缩压 (SBP)、平均动脉压 (MAP)、舒张压 (DBP)、心率 (HR)及呼吸 ,测定血浆 ET、CGRP的含量。结果 组内比较 :B组各时点血压、心率无差异 (P<0 .0 5 ) ;C组随时间的延长 ,2 0 min、 30 min的 SBP、MAP、DBP、HR均下降 ,尤以 30 m in为著 (P<0 .0 5或 0 .0 1)。组间与 A组比较 :各组基础值无差异 ;B组仅在 2 min、10 min时 HR下降 (P<0 .0 5 ) ;而 C组 SBP、MAP、DBP、HR均下降 (P<0 .0 5或 0 .0 1) ,且 HR随时间的延长与 A组的差异明显增加 ,30 min时 P<0 .0 0 1;B组和 C组血浆中 CGRP增高、ET下降 ,CGRP/ ET比例增高 ,C组改变显著。结论 异丙酚促进 CGRP释放 ,抑制 ET的释放 ,通过改变 CGRP/ ET的平衡呈时间剂量依赖性降血压 。
Objective To investigate the effects of propofol on hemodynamics a well as Endothelin and Calcitonin Gene Related Petide(CGRP)of plasma in rats.Methods:Twenty four Wistar rats at age of 12~14 weeks were randomly allocated into 3 groups.Group A:control(n=8,only injected with 0.9% NaCl);in other two groups:anesthesia was induced with intravenous propofol 5mg·kg -1 in volume 1 2ml·kg -1 ·h -1 for 30min,respectively.Systolic blood pressure(SBP),mean blood pressure(MBP),diastolic blood pressure(DBP) and heart rate(HR) were monitored continously.After 30min infusion,the concentrations of Etand CGRP of the plasma were determined.Results In the same group:there were no significant differences of blood pressure and HR in group B;In group C,SBP,MBP,HR decreased significantly at 20min,especially at 30min(P< 0.05 or 0.01).As compared with those in group A:There were no significant differences in baselines among three groups;The HR at 2min and 10min in group B decreased with significant difference(P<0 05).The SBP,MBP,DBP and HR reduced greatly in group C(P<0 05 or 0 01),among which HR decreased markedly as the infusion time went by,P<0.001 at 30min.The level of CGRP of plasma increased,ET decreased and ratio of CGRP to ET was also enhanced in both group B and C.But the changes in group C were greater than those in group B.Conclusions propofol facilitates the release of CGRP,but inhibits the release of ET from endothelial cells.Propofol reduces blood pressure and slows down heart rate by changing the balance of CGRP/ET in dose and time dependent manner.
